Genes affected by dielectrophoretic manipulation of living cells in micro-fluidic devices

Abstract

A complementary DNA microarray multiplex technology has been used to identify the gene expression changes due to dielectrophoresis (DEP) manipulation of living cells in microdevices. Alternations in 6754 genes have been studied. The effects of both phenomena positive DEP (pDEP) and negative DEP (nDEP) on cells have been investigated. Cells were kept in microdevice for one-hour under the influence of the electric field used in DEP. Gene modifications of three specimens (pDEP, nDEP, and control) were compared. The effect of 20V peak-to-peak AC on living cells has been found to affect to certain extent cells growth and proliferation. Moreover, no effect on cells viability was found. MDA231 cells suspended in low conductivity medium were kept in electric field of same intensity as the one used in DEP. Cells were found capable to grow and multiply after exposure. Changes in genes expression were investigated using microarray analysis. As result, the exposure of living cells to 10 kHz frequency AC field was found to alter 75% of the genes compared to only 24% in case of 100 kHz frequency.
